Effects of adjuvant propofol on antitumoral effects of carboplatin in experimental endometrial cancer: possible epigenetic associations

Endometrial cancer (EC) is a gynecological malignancy classified into two biologically distinct subtypes, Type I and Type II EC. The more aggressive Type II EC exhibits a poor response to conventional platinum-based chemotherapy. Propofol, an anesthetic agent in gynecologic surgery, has been reported to exert antitumor effects beyond anesthesia; however, its role as a chemoadjuvant in EC remains insufficiently explored. In this study, we investigated the effects of propofol combined with carboplatin on drug resistance- and metastasis-associated microRNAs and genes using in vitro and in vivo Type II EC model. Propofol treatment significantly reduced cancer cell viability and migratory capacity, while the combination treatment showed greater antimetastatic effects compared with carboplatin alone. In xenograft models, combination therapy resulted in a significant reduction in tumor volume and suggested reduced metastatic involvement in lung and liver tissues. Molecular analyses revealed downregulation of oncogenic microRNAs (miR-21, miR-135a) and upregulation of tumor-suppressive microRNAs (miR-34b, miR-98), accompanied by reduced mTOR, c-Myc, MRP7, and N-cadherin expression and increased PTEN and HMGB1 levels. Collectively, these findings indicate that propofol is associated with greater antitumor effects when combined with carboplatin, accompanied by changes in miRNA-gene expression patterns.

Stress distribution in titanium and zirconia implants with full-arch prostheses: a finite element analysis

Objective: The stress distribution and mechanical properties of materials used in dental implants are critically important for implant success and long-term stability. The aim of this study was to evaluate the stress distribution on implant, prosthetic system, and peri-implant tissues when titanium and zirconia implants are used with different framework materials using three-dimensional finite element analysis (FEA). Methods: A three-dimensional edentulous maxilla model was reconstructed from computed tomography data, and twelve finite element models were subsequently developed based on this anatomy to represent different implant and framework configurations. Within the scope of the study, titanium and zirconia implants were modeled in combination with cantilevered and noncantilevered prosthesis types and Co-Cr, PEKK, and zirconium framework materials. An oblique static load of 200 N was applied at a 45° angle in the palato-buccal direction to the central fossa of the first molar on the models. The Von Mises and principal stress distributions in the implant, abutment, screw, framework, and peri-implant bone were examined. Results: The highest Von Mises value was recorded in the cantilevered PEKK framework zirconia implant model (378.388 MPa). Compared with the Co-Cr and zirconium framework, the PEKK framework resulted in higher Von Mises stresses and greater prosthesis displacement. The presence of a cantilever increased stress levels in all framework materials, and stress concentrations were observed particularly in posterior abutments and peri-implant bone. Furthermore, stress values in trabecular bone were recorded above the physiological threshold level in the cantilevered PEKK framework models. Conclusions: Although zirconia implants exhibited greater stress than titanium implants, they remained within safe limits. Rigid framework materials and cantilever-free designs provide more balanced biomechanical results.

The Role of Boron in the Diet and as a Dietary Supplement

Boron is a naturally occurring trace element that has historically received limited attention in human nutrition and is of increasing biological importance. It is widely found in nature, primarily through food and drinking water, and has attracted attention for its potential health benefits. Emerging evidence suggests that boron supplementation may support bone health, cognitive function, mineral and hormonal metabolism, the antioxidant defense system, body weight regulation, and inflammation reduction. This review examines the physiological roles, health, and clinical effects of boron, both through dietary intake and supplementation. Although not currently classified as an essential nutrient, boron appears to be biologically important, and it is thought that supplementation may have positive effects on many health conditions, including osteoporosis, osteoarthritis, body weight regulation, cancer prevention and treatment, diseases associated with oxidative stress, neurological and mental disorders, and hormonal imbalance.

The Comorbidity of Specific Learning Disorders in Attention Deficit Hyperactivity Disorder

Attention deficit hyperactivity disorder (ADHD) is a neurobiological disorder that begins in childhood, with symptoms continuing throughout life, negatively impacting an individual’s cognitive, social, and academic functioning. Specific learning disability (SLD) is a developmental disorder characterized by its initial signs being noticeable during the preschool years. The broad category of SLD encompasses various skill deficits, but the most common symptoms are typically divided into three broad subcategories: reading disorder, written-language disorder, and mathematics disorder. Generally, there is an estimated comorbidity rate between SLD and ADHD ranging from 31% to 45%. While ADHD is conventionally defined as an externalizing disorder, children with this condition often exhibit learning difficulties and academic problems in addition to behavioral issues such as impulsivity, oppositional behavior, and conduct problems. These academic difficulties are frequently overlooked, as behavioral problems, often linked to impulsivity, tend to take precedence. The purpose of this review is to draw attention to these overlooked areas of concern in children diagnosed with ADHD, both in clinical practice and academic literature, and to review relevant literature studies.

Spatial characterization of the phoenixin receptor in the lacrimal and major salivary glands

Objective: To identify and characterize the expression of phoenixin receptor GPR173 (G protein-coupled receptor 173) in the rat lacrimal and major salivary glands (parotid, submandibular, and sublingual). GPR173 is associated with hormonal regulation and autoimmunity; however, its expression in exocrine glands has yet to be characterized. Design: Under anesthesia, eight Sprague Dawley rats underwent bilateral dissection of the lacrimal and major salivary glands. GPR173 expression was assessed using Western blotting and immunofluorescence. Immunoreactivity was quantified using ImageJ software and compared using the Friedman test with DunnBonferroni post hoc analysis. Results: GPR173 protein expression was found in all examined exocrine glands. In the major salivary glands, GPR173 was detected in both acinar and ductal cells, while expression was confined to acinar cells in the lacrimal gland. The submandibular and parotid glands showed significantly higher immunoreactivity than the lacrimal gland (p < .01 and p < .05, respectively). Ductal epithelium showed stronger immunopositivity compared to acinar epithelium. GPR173 immunoreactivity was also observed in periductal stromal tissue in major salivary glands. Conclusion: GPR173 expression was observed in both acinar and ductal cells of all major salivary glands, while in the lacrimal gland, it was restricted to the acinar cells. This differential localization suggests tissue-specific relevance.
